This is somewhere in the neighborhood of 10 billion for a single mission. This is never going to happen since it would be following a much better plan.
Human missions to deep space destinations require large amounts of mass in LEO because we need to protect and preserve the lives of the crew, take all consumables and fuel for the journey, and carry the equipment and supporting machines needed to carry out the mission objectives.No. We only need enough mass to get the job done. More mass is more cost. Most equipment should be sent ahead and waiting. Nothing need be stationed in LEO.
No existing commercial launch vehicle (nor any anticipated in the near future) has the launch capacity of the SLS.The SLS doesn't have any launch capacity yet. Other vehicles with greater capacity could actually become reality sooner.

Dragon 2 exists and can land anywhere including on mars. FH is made from existing cores and will be flying soon. It has been delayed by other priorities but that situation has drawn to a close.

Falcon Heavy, per Musk, isn't going to have crossfeed. However, even without crossfeed, the cores don't hit MECO at the same time; the center core operates at a lower throttle setting, so the side cores deplete first.
To me, the most preposterous part of Spudis's argument is this;
"with fewer launches of larger payloads, a lower launch cost per unit mass is realized."
No, it isn't. It's dollars per pound, and thus based on the vehicle's cost per pound ratio. Of course you don't want to try assembling 50 pieces, but simply doubling (at worst) the dockings needed is not a concern.
I also like they way he dismissed Falcon Heavy due to it not having flown yet (it's months from flying) while pretending SLS is existing; SLS is a lot further from flying than Falcon Heavy, and won't even see its first flight until 2020 at the earliest. I also note that he's using outdated figures for Falcon Heavy payload capacity.
SLS does minimize the orbital docking issue though; because of the cost of SLS, you won't have money for any payloads, so no docking at all.
